          Statins have been developed as a medicine for reducing serum cholesterol by inhibiting the rate-limiting enzyme, hydroxymethylglutary-coenzyme (HMG-CoA) reductase. Studies over the past 10 years have recognized them as useful medicine to ameliorate neurodegenerative disorders, such as cerebral ischemic stroke, vascular, Alzheimer’s disease, and Parkinson’s disease, although the underlying neuroprotective mechanisms are not well understood. Our recent studies have shown that simvastatin can modulate dopaminergic, glutamatergic (NMDA), histaminergic, and acetylcholine muscarinic neurotransmissions. Using an experimental Parkinsonia rat model, we investigate the roles of simvastatin in improving locomotor activity, anxiolytic-like behaviours and long-term memory. Consistently, simvastatin is found able to prevent dopaminergic neurodegeneration, to reverse the downregulation of NMDA and muscarinic M1/4 receptors. These findings contribute to a better understanding of simvastatin’s neuroprotective effects in treating neurodegenerative disorders through the modulation of neurotransmissions.
     
          专家简介:
          澳大利亚University of Wollongong抗精神药物研究室主任,副教授。1983年毕业于西南师范大学生物系,1998年获得澳大利亚University of New England神经科学博士学位,1998年-2003年先后在澳大利亚University of New England、University of New South Wales从事博士后研究,2003-今在澳大利亚University of Wollongong担任研究员、高级研究员、副教授。主要从事神经性疾病的分子机理与抗精神药物的作用靶标和新药研究,目前承担抗精神病药物机制与副反应和新一代抗精神药物等澳大利亚国家NH&MRC基金课题三项,发表SCI论文60余篇。目前担任国际上多家跨国制药企业科学顾问和研究专家、多个国际杂志的审稿专家。
